ABSTRACT

Neuroplastin 65 (Np65) is an immunoglobulin superfamily cell adhesion molecule involved in synaptic formation and plasticity. Our recent study showed that Np65-knockout (KO) mice exhibit abnormal cognition and emotional disorders. However, the underlying mechanisms remain unclear. In this study, we found 588 differentially-expressed genes in Np65-KO mice by microarray analysis. RT-PCR analysis also revealed the altered expression of genes associated with development and synaptic structure, such as Cdh1, Htr3a, and Kcnj9. In addition, the expression of Wnt-3, a Wnt protein involved in development, was decreased in Np65-KO mice as evidenced by western blotting. Surprisingly, MRI and DAPI staining showed a significant reduction in the lateral ventricular volume of Np65-KO mice. Together, these findings suggest that ablation of Np65 influences gene expression, which may contribute to abnormal brain development. These results provide clues to the mechanisms underlying the altered brain functions of Np65-deficient mice.

ABSTRACT

OBJECTIVE:To study the effect of paroxetine combined with low dose of olanzapine on sleep process and architec-ture of depression patients with insomnia. METHODS:84 depression patients with insomnia were randomly divided into control group and observation group. Control group was given 20 mg Paroxetine tablet, once every morning;observation group was addi-tionally given 2.5 mg Olanzapine tablet, once before going to bed. Sleep quality [Pittsburgh Sleep Quality Index(PSQI)],depres-sion scores [Hamilton Depression Scale (HAMD)],sleep process [sleep latency (SL),awakening times (AT),the actual total sleep time (TST),sleep efficiency (SE),rapid eye movement (REM) sleep latency (RL)] and sleep architecture [sleep stage 1 (S1),2(S2),3(S3)and the proportion of REM to sleep] in 2 groups before and 3,6 months after treatment and the incidence of adverse reactions(ADR)were observed. RESULTS:After treatment,PSQI and HAMD scores in 2 groups were significantly lower than before, and gradually decreased by time, and observation group was lower than control group;TST in observation group was significantly higher than before and control group,S1 in observation group was significantly lower than before,SE,S3 and REM in 2 groups were significantly higher than before, and observation group was higher than control group,SL,AT,RL and S2 were significantly lower than before, and observation group was lower than control group, the differences were statistically significant (P<0.05). There were no obvious ADR in 2 groups. CONCLUSIONS:Paroxetine combined with low dose of olanzapine can sig-nificantly relieve depression, optimize sleep process and sleep architecture, then impove sleep quality.

ABSTRACT

OBJECTIVETo investigate the application of risk grading and classification for occupational hazards in risk management for a shipbuilding project.

METHODSThe risk management for this shipbuilding project was performed by a comprehensive application of MES evaluation, quality assessment of occupational health management, and risk grading and classification for occupational hazards, through the methods of occupational health survey, occupational health testing, and occupational health examinations.

RESULTSThe results of MES evaluation showed that the risk of occupational hazards in this project was grade 3, which was considered as significant risk; Q value calculated by quality assessment of occupational health management was 0.52, which was considered to be unqualified; the comprehensive evaluation with these two methods showed that the integrated risk rating for this shipbuilding project was class D, and follow- up and rectification were needed with a focus on the improvement in health management.

CONCLUSIONThe application of MES evaluation and quality assessment of occupational health management in risk management for occupational hazards can achieve objective and reasonable conclusions and has good applicability.

ABSTRACT

Objective To investigate the expression and relationship of Jab1 and P27kip1 in ovarian cancer cell HO-8910. Methods Cells were treated with serum starvation and release. The expression and distribution of Jab1 and P27kip1 were detected by western blot and subcellular fractionation.HO-8910 cells were transfected in vitro with pcDNA3.1-MYC-Jab1.Western blot as well as subcellular fractionation was used to detect the expression and localization of P27kip1.Results The growth of HO-8910 cells was blocked by serum starvation.P27kip1 increased while Jab1 decreased. The reverse changes were found after serum release.P27kip1 and Jab1 could form compound in HO-8910 cells detected by immunoprecipitation.48h after transfected by pcDNA3.1-MYC-Jab1,the expression of P27kip1 decreased and the distribution of P27kip1 translocated from nucleus into cytoplasma in HO-8910.Conclusion Jab1 may control the location and expression of P27kip1 through integrating with P27kip1,and participate in regulating the growth of ovarian cancer cells through interfering with the function of P27kip1.

ABSTRACT

Objective:To observe the effect of Radix Paeoniae Rubra on intimal proliferation, activating of angiotensin Ⅱ(AgⅡ) and expression of Mitogen-activated protein kinase phosphatase-1(MKP-1) after carotid artery balloon injury in cholesterol-fed rabbits. Methods: Male rabbits were randomly divided into Radix Paeoniae Rubra(PPR) groups: high dose group(75、50、25g.kg-1.d-1;n=8),middle dose group(2g.kg-1.d-1),low dose group(1g.kg-1.d-1;n=8) and control group. Both groups received high fat forage(2% cholesterol + 5% lard).Balloon injury of carotid artery was performed.Carotid artery were harvested at the end of 10 weeks.Expression level of AgⅡwas measured by radioimmunoassay.MKP-1 expression was determined by RT-PCR.Immunohistochemical staining and morphological detection were adopted. Results: Compare with control group, expression of AgⅡ decreased obviously(P
